1.PBS 緩沖液（pH7.4，實驗室常用的10mM磷酸緩沖鹽溶液（1X））

1.細胞處理需要小心操作,盡量避免人為的損傷細胞。
2.使用含EDTA的細胞消化液。
3.洗滌細胞離心轉速不可超過800×g。
4.乙醇固定時離心大概采用2000×g力5分鐘，離心力太小可能部分細胞難以沉淀。? 5.固定時在振蕩器上輕輕振蕩細胞，并緩慢滴加75%乙醇，直接加入會導致細胞團聚的現象，很難重懸成單細胞。或者先用冷PBS懸浮細胞，充分懸浮，使細胞充分分散成單細胞。之后緩慢加入無水乙醇，終濃度為70-90%乙醇。乙醇固定之后須無細胞沉淀。

1. 收集樣本細胞，細胞數量在1-5*106個。
2. 用冷PBS洗滌細胞兩次。
3. 在15ml離心管中，用500 μl PBS重懸細胞。
4. 慢慢滴加冷的無水乙醇或95%乙醇至終濃度為70%-80％冰凍乙醇。-20℃固定1小時或4℃固定過夜。
5. 在1000g離心10分鐘，棄上清，收集細胞。
6. 用冷PBS洗滌細胞一次。
7. 用500 μl冷PBS重懸細胞。
8. 加入Rnase A溶液20 μl， 37℃水浴30分鐘。
9. 400目篩網過濾。
10. 在1000g離心10分鐘，棄上清，收集細胞。
11. 加入400 μl PI染液重懸細胞，輕輕混勻后4℃避光孵育30分鐘-1小時。
12. 流式檢測結果。激發波長為488nm。采用適當分析軟件進行細胞DNA含量分析和光散射分析。

1.染色后找不到細胞？
固定細胞可能難以離心沉淀；固定細胞可能成團；固定細胞膜容易損傷成碎片。操作過程中注意以上幾點。

2.可以不固定細胞嗎？
本試劑盒可以用于非固定細胞的周期檢測。
